摘    要:目的]分析桉树组织培养中的“玻璃化”现象发生的原因,探索克服或减少“玻璃化”现象的措施。方法]以巨桉品种广林9为试材,基本培养基为:琼脂3.0‰,糖30‰、NH4+5.0ml、Ca2+10ml、水为自来水,针对组织培养中出现“玻璃化”现象设11个配方处理,研究改变培养基中琼脂、蔗糖、NH4+和Ca2+含量以及水质情况对克服桉树组织培养中的“玻璃化”的影响。结果1研究提出一些克服或减少桉树组织培养中玻璃化现象的措施:配制培养基时固化剂的合适用量一般在3.5%。;培养基中蔗糖含量可适当增加10‰左右。以降低培养基的渗透势,阻逼培养材料过量吸收水分;培养基中Ca含量可增加1.5ml、NH4+含量可降低2.0moL/L;煮制培养基时最好用自来水。结论]该研究为克服桉树组织培养中“玻璃化”现象提供科学依据。

Abstract:Objective] The research aimed to analyze the generated cause of vitrification phenomenon in tissue culture of eucalyptus,and explore the measures that would overcome or lessen vitrification phenomenon.Method] With Guanglin 9 of eucalyptus grandis variety as tested material,the basic medium was: agar 3.0‰,sugar 30‰,NH+4 5.0 ml,Ca2+ 10 ml,and water was tap water,11 formula treatments were set up aiming at the vitrification phenomenon occurred in tissue culture to study the effect of changing the contents of agar,sucrose,NH+4 and Ca2+ and the water quality state in medium on conquering the vitrification in eucalyptus tissue culture.Result] Some measures of overcoming or decreasing vitrification phenomenon in tissue culture of eucalyptus were proposed in the research as follows: generally,the suitable dosage of hardener was 3.5‰ when preparing medium;the sucrose content in medium could be increased about 10‰ properly to reduce the osmotic potential of medium and prevent culture material from absorbing moisture excessively;the Ca content in medium could be increased by 1.5 ml,while NH+4 content could be decreased by 2.0 mol/L;its better to use running water when cooking medium.Conclusion] The study supplied scientific basis for overcoming vitrification phenomenon in tissue culture of eucalyptus.
