鹌鹑新城疫病毒分离株F基因的克隆和序列分析

摘    要:研究以新城疫鹌鹑分离株(LA005)的基因组RNA为模板,应用RT-PCR技术扩增出其F基因的主要功能区片段,并进行了克隆和序列分析。结果表明:所扩增的目的片段长度为813bp,裂解位点氨基酸序列为112R-T-Q-R-R-Fll7,和F48E9标准强毒株的核苷酸和氨基酸的同源率分别为98.9%和98.5%,Cys残基位点和糖基化位点的数目和位置完全一致,说明该分离毒是一株和F48E9标准强毒株亲缘关系很近的强毒株。

Abstract:Main function segment of F gene was amplified by RT-PCR using genomic RNA template of quail NDV isolate, and cloned into PMD 18-T vector.The sequence analysis showed that the length of the F gene segment was 813bp . The amino acid sequence of the cleavage site region was 112R-R-Q-R-R-F 117. Maching to F48E9 strain, the sequence homologous rates of nucleotide and amino acids were 98.9% and 98.5% respectively, the numbers and position of cysteine residues and glycosylation sites were sameness at all. These indicated that the quail NDV isolate belonged to the virulent NDV stvains, and similar to F48E9 strain in heredity.
