高效毛细管电泳法同时分离氟喹诺酮类 与四环素类药物的研究

摘    要:采用高效毛细管电泳建立4种氟喹诺酮类(FQS)与2种四环素类(TC)药物同时分离的方法。通过考虑缓冲液离子浓度、pH、有机添加剂以及分离电压、温度等电泳参数,通过正交试验确立了电泳的最佳条件:紫外检测波长270 nm,缓冲溶液为50 mmol.L-1硼砂-200 mmol.L-1硼酸,pH 8.41,分离电压为18 kV。结果表明,6种药物在12 min内达到完全分离,且各组分浓度与峰面积呈良好的线性关系,其相关系数R2为0.998 2~1,迁移时间的相对标准偏差为0.25%~0.68%,峰面积相对标准偏差为7.3%~9.8%。

Abstract:A method for the simultaneous separation of fluoroquinolones(FQs)and tetracyclines(TCs)by high performance capillary electrophoresis(HPCE) was developed.The effects of several factors such as the con-centration and pH of running buffer,the organic added to the electrolyte,the voltage and the temperature on sepa-ration were investigated.The optimal conditions were as follows.The UV detection wavelength was 262 nm.The running butter consisted of 50 mmol·L-1 NaB4O7-200 mmol·L-1 H3BO3 with pH 8.41,and the separation voltage was at 18 kV.The results showed that the six remedies can be well separated within 12 min.Each had a good lin-ear relationship(R2=0.9982~1).The RSD was 0.25%~0.68% and 7.3%~9.8%.
