摘    要:以树兰(Epidendrum)为材料,分别研究了NaClO和HgCl2对树兰侧芽、茎段和花梗3种外植体的消毒效果,探讨树兰3种外植体的有效消毒方法。试验结果表明:(1)3种外植体中最易消毒的为花梗,其次是侧芽,最难消毒的为茎段;(2)HgCl2对3种外植体的消毒效果优于NaClO;(3)3种外植体最适宜的消毒方法分别为:花梗0.1%HgCl2消毒8 min,侧芽0.1%HgCl2消毒4 min,茎段0.1%HgCl2消毒10 min,其存活率分别为90.9%、42.3%和30.9%。本研究结果将为树兰外植体途径的组培再生提供理论依据和技术指导。

Abstract:To study the effective sterilization method of Epidendrum, the disinfection effect of NaClO and HgC12 for Epidendrum lateral bud, stem and flower stalk were analyzed respectively. The results showed that: (1) Flower stalk was the most easily disinfected among the three explants, then was lateral bud, the other was stem. (2) The disinfection effect of HgCl2 was apparently better than NaClO. (3) The optimum sterilization method of flower stalk, lateral bud and stem of Epide ndrum was disinfected with 0.1% HgCl2 for 8 min, 4 min and 10 min, respectively, and the survival rate accordingly reached up to 90.9%, 42.3% and 30.9%. This result will provide theoretical basis and technical guidance for tissue culture of Epidendrum.
