摘    要:通过农杆菌浸染的转基因技术获得拟南芥转基因株系,亚细胞定位研究发现AtCYP1在细胞膜和细胞核中有明显表达。利用过表达技术,对转基因过表达、共抑制株系进行研究,结果表明,AtCYP1转基因共抑制株系在开花时间和抽苔时间均较野生型晚2~3 d且抽苔前叶片始终多于对照,而过表达株系则相反,说明AtCYP1基因参与了拟南芥开花时间的调控。另外,悬浮细胞培养表明,AtCYP1基因的过量表达提高了细胞的最高相对增长率,共抑制则反之,生长周期也发生了变化,说明AtCYP1基因对拟南芥悬浮细胞的增殖有调控作用。

Sub-cellular Localization and Physiological Functional Analysis of At- CYP 1 in Arab idops is thaliana

Abstract:In order to understand physiological function of AtCYP1, we obtained Arabidopsis transgenic lines by Agrobacterium-mediated transformation. It was found by subcellular localization studies that AtCYP1 was ex- pressed mainly in nucleus and on membrane. While we also characterized the transgenic over-expressed and co-in- hibit lines through gene over-expression technology. The results showed that AtCYP1 transgenic AtCYP1 co-in- hibited homozygous lines postpone flowering time later 2 or 3 days later than that of wild-type plant, and their rosette leaves at bolting stage produce more than that of wild-type (blank control) and negative control (empty vec- tor). Whereas the transgenic AtCYP1 over-expression lines exhibit complete contrary phenotypes. Therefore we deduced that AtCYPI might be involved in the regulation of flowering time. Additional, the result from cell sus- pension culture showed that over-expressed AtCYP1 gene will enhance the maximal cell proliferation rate as well as change the growth period, but the co-inhibited homozygous lines did contrary, which would imply that AtCYP 1 could involve in the regulation of cell proliferation pathway.
