百合科十二卷属玉露的组培快繁关键技术研究

摘    要:为建立十二卷属多肉花卉玉露的快繁技术体系,以当年抽生的花序为外植体,以MS为基本培养基,研究不同的激素配比对外植体的诱导、分化、增殖、壮苗与生根的影响,建立适合玉露离体快繁的技术体系,最佳启动培养基为MS 6-BA 2.0 mg/L NAA 0.02 mg/L,愈伤组织诱导及分化培养基MS 6-BA 0.5 mg/L KT 0.5 mg/L NAA 0.01 mg/L,愈伤组织诱导率达87.1%,丛芽分化率达52.3%,生根培养基为1/2MS IBA 0.5 mg/L NAA 0.1 mg/L AC 0.5 g/L。试管苗生根率达97.5%,炼苗成活率达90%以上。本项研究结果可以在不损伤母本的基础上实现玉露的人工组培快繁,通过此项技术可以实现玉露种苗的规模化生产。

Key Techniques for Rapid Propagation of Haworthia cooperi var. pilfera of Liliaceae

Abstract:The research aims to establish a rapid regeneration system of Haworthia cooperi var. pilfera. The authors used the inflorescence as explants and MS as the basic medium to research the effect of different proportions of hormones on inducing differentiation, growth, expanding propagation, strong young plants and producing roots. The efficient technique system for rapid propagation was established. The result showed that the optimal priming medium was MS 6-BA 2.0 mg/L NAA 0.02 mg/L and the optimal induction and differentiation medium was MS 6-BA 0.5 mg/L KT 0.5 mg/L NAA 0.01 mg/L and the callus induction rate was 87.1%, the bud differentiation rate reached 52.3%. The best rooting medium was 1/2MS IBA 0.5 mg/L NAA 0.1 mg/L AC 0.5 g/L. The root growing rate of tube-seeding was 97.5% and the transplanted seedling had a survival rate up to 90%. The techniques based on this research for tissue culture and rapid propagation had no damage to the female and could be applied to large-scale production of Haworthia cooperi var. pilfera seedlings.
