摘    要:为建立槐枝中高丽槐素含量的检测方法,以自制高丽槐素为对照品,采用Diamonsil C18色谱柱(250mm×4.6mm,5μm)为固定相,以乙腈-0.1%冰醋酸水溶液(40∶60)为流动相,流速1.0mL/min,检测波长310nm,柱温30℃,进样量为20μL,外标法定量检测贵州6个不同产地(贵州省贵阳市观山湖区、南明区龙洞堡、云岩区东山、白云区都拉营、花溪区小河、贵州省遵义市)槐枝的高丽槐素含量。结果表明:在该色谱条件下,高丽槐素进样浓度为1.328~39.8μg/mL时与峰面积呈良好的线性关系(R2=0.999 6),平均回收率为102.37%,RSD为1.59%。建立的高效液相色谱法(HPLC)测定槐枝中高丽槐素含量,其方法操作简便、准确,重复性和稳定性好。南明区龙洞堡槐枝的高丽槐素含量最高,达100.56μg/g;其他几个产地的含量差异不大,在57.56~79.9μg/g。

Determination of Maackiain in Sophora japonica by HPLC
Abstract:In order to determine the content of maackiain in S.japonica,a new high performance liquid chromatography (HPLC) method was established,taking home-made maackiain in laboratory as the reference sample.Diamonsil C18 column (250 mm× 4.6 mm,5μm) was used as stationary phase,ACN-0.1% HAC (40 ∶ 60) with a gradient elution was used as the mobile phase.The column temperature was 25C,the flow rate was 1.0 mL/min,the UV detective wavelength was 310 nm,the injection volume was 20 μL,and the sample was quantified with the external standard method (ESTD).Through the quantified test of six different regions samples of maackiain under the above chromatography conditions.Under these conditions a good linear relationship was obtained with peak area when the sample concentration was 1.328~39.8 ig/mL(R2 =0.999 6),and the average recovery was 102.37% with RSD of 1.59%.In a word,HPLC method established which was used to test the content of maackiain in S.japonica have a series of advantages,such as convenient operations,accuracy,high repeatability and good stability.The content of maackiain in S.japonica sampled in Nanming District reached the highest,100.56 μg/g,which were of little difference in S.japonica in other producing areas,ranging from 57.56 to 79.9 μg/g.
