悬浮培养及培养条件对灵菊七细胞中可溶性多糖合成的影响

摘    要:以灵菊七叶片诱导的愈伤组织为材料进行悬浮培养,研究培养条件对愈伤细胞、细胞内可溶性多糖合成的影响。结果表明,在培养基MS+0.5 mg/L BA+0.5 mg/L NAA+1.0 mg/L GA中悬浮细胞表现出对数生长期最长、可溶性多糖含量最高的特点;离子交换层析纯化结果显示,悬浮细胞中可溶性多糖分了与植株中含有相同的3种主要多糖。培养基MS+0.5 mg/L BA+0.5 mg/L NAA+1.0 mg/L GA,促进可溶性多糖的生物合成,最适于灵菊七愈伤细胞的悬浮培养。本研究初步建立了灵菊七愈伤细胞悬浮培养快速获取可溶多糖的方法,为深入研究灵菊七多糖药理活性奠定了基础。

Suspension Culture of Gynura medica and Effect on Bio-Synthesis of Soluble Polysaccharides by Culture Conditions

Abstract:Callus induced from leaf of Gynura medica was cultured and regenerated, then inoculated in 4 liquid medium to study effect on callus and bio-synthesis of soluble polysaccharides by culture conditions. The result showed, when the callus was cultured in MS+0.5 mg/L BA+0.5 mg/L NAA+I.O mg/L GA, that suspen- sion cell exhibited characteristics with the longed logarithmic growth phase and the highest content of soluble polysaccharides. Ion-exchange result showed that suspension cell contained the same 3 primary soluble polysac- charide compared with plant of G. medica. It was improved that the media of MS+0.5 mg/L BA+0.5 mg/L NAA+I.O mg/L GA was optimum media and promotes bio-synthesis of soluble polysaccharides for suspension cell. It was concluded that study established the method for obtaining G. medica soluble polysaccharides by suspension culture of callus, and established the basic work for study on pharmacology of G. medica soluble polysaccharides.
