细叶连翘的组织培养与快速繁殖

摘    要:[目的]研究细叶连翘组织培养与无性快繁技术。[方法]以细叶连翘的茎及茎尖为外植体,以MS为基本培养基,添加不同浓度的6-BA和IBA,配制不同的培养基,对细叶连翘试管苗进行分化与诱导、增殖与生根培养试验,并对试管苗进行炼苗与移栽。[结果]经分化与诱导培养后的无毒茎尖分化苗接种到增殖培养基中,10d后分化出丛生芽,20d后苗高3~4cm,将3~4cm的丛生苗转入生根培养基中,7d后有不定根形成,20d后丛生苗生根率达100%,根长2~3cm的幼苗经炼苗和移栽后,成活率可达98%。[结论]获得了细叶连翘茎尖培养的启动培养基(MS+6-BA0.5mg/L+IBA0.2mg/L)、增殖培养基(MS+6-BA1.5mg/L+IBA0.2mg/L)和生根培养基(1/2MS+IBA0.5ms/L),建立了细叶连翘的组织培养与快速繁殖体系。
