海南黑山羊CAST基因的时空表达分析

摘    要:采用SYBR Green实时荧光定量PCR技术检测了CAST基因在1、2、3、4月龄和12月龄海南黑山羊骨骼肌、心肌、肝脏和脂肪组织中mRNA表达差异和发育规律,为揭示CAST基因的生物学功能及其对肉质的表达调控机理提供参考依据.结果表明,CAST基因在未成年羊的骨骼肌中mRNA表达量极显著高于其他组织(P<0.01),且在2月龄时达到最高水平;成年后在心肌、肝脏和脂肪中的表达量极显著升高(P<0.01),在骨骼肌中的表达量次之,但在脂肪中的表达量始终相对较低;该基因在3、4月龄各组织中的mRNA表达量相对较低,且四种组织在3、4月龄间表达差异不显著(P>0.05).

Abstract:The mRNA expression differences and developmental changes of Calpastatin (CAST) gene in the skeletal muscle, heart, liver, and adipose tissue of Hainan Black Goat from the 1, 2, 3, 4 months and 12 months of age were investigated by SYBR Green real-time PCR technique to explore the biological func- tion of CAST gene and its expression and regulation mechanism of meat. The results showed that: the CAST gene mRNA expression in skeletal muscle of lambs was significantly higher than other tissues (P〈 0.01), and reached its highest level at 2 months of age. The expression level in heart, liver, and adipose tissue of adults significantly increased (P〈0.01), and the expression in skeletal muscle was the second, but its expression level in adipose tissue was always lower,in addition, the CAST mRNA gene expression of the four tissues in 3 and 4 months was relatively lower, and there was no significant expression differ- ence between the two months (P〉0.05).
