黑番鸭VIPR-1基因第12内含子SNP与就巢性状关联分析

摘    要:VIPR-1基因是研究家禽就巢性状的重要候选基因,以就巢期黑番鸭和非就巢期黑番鸭为研究对象,通过PCR-RFLP和测序的方法,对VIPR-1基因内含子12进行多态性检测,并进行相关性分析.结果表明,VIPR-1基因内含子12存在一个StuI酶切位点,该位点由于在内含子988 bp处发生了A→G的碱基突变,产生了AA、AG、GG三种基因型.其中就巢黑番鸭只表现为AG(1.0)一种基因型,而非就巢黑番鸭表现为AG(0.4167)、AA(0.1666)、GG(0.4167)三种基因型,就巢期黑番鸭与非就巢期黑番鸭两者之间基因型的分布差异极显著(P<0.01).试验表明黑番鸭VIPR-1基因第12内含子A988G突变与黑番鸭的就巢性状显著相关,为利用分子遗传标记辅助选择来降低或消除黑番鸭的就巢性,最大程度上提高黑番鸭的产蛋性能提供了新的依据.

Abstract:As a critical candidate gene of poultry broodiness,intron 12 of the vasoactive intestinal peptide-1 gene of black Muscovy ducks was analyzed by PCR-RFLP.The results indicated that it was a site of StuI digestion with intron 12 of VIPR-1 gene.An A→G mutation at the site 988 bp of this gene was detected by DNA sequencing which produced three sorts of genotypes AA,AG and GG.The broodiness of black muscovy ducks showed only one genotype AG (1.0),while the non-broodiness black muscovy ducks showed three genotypes of AG (0.4167),AA (0.1666),GG (0.4167).By chi-square test,there were significant differences (P〈0.01) between broodiness black muscovy ducks and non-broodiness black muscovy ducks in genotypes,which indicated that A988G mutation in intron 12 of VIPR-1 gene might reduce or eliminate the broodiness traits of ducks and improve the performance of black Muscovy duck's egg.
