摘    要:本文对甜菜线粒体DNA的提取方法进行了改进,在不需密度梯度离心条件下即可获得高纯度的DNA。优化了甜菜线粒体的RAPD的反应条件,对细胞质雄性不育甜菜的不育系及其保持系的线粒体DNA(mtDNA)进行了RAPD(随机扩增多态DNA,randomly amplified polymorphic DNA)分析,结果表明:甜菜细胞质雄性不育系和保持系线粒体DNA之间存在丰富的多态性。

Abstract:Mitochondrial DNA (mtDNA) extracted from cytoplasmic male sterile(CMS) line and its maintainer line of sugarbeet was studied by means of random amplified polymorphic DNAs(RAPD). The fingerprints of the amplification by polymerase chain reaction (PCR) of random DNA segments was analysised. The results indicate that there are the obvirous difference between CMS line and maintainer line revealed by RAPD technique. Meanwhile this test improved the extraction method of mtDNA also optimized the program of RAPD.
