绵羊雌激素受体基因外显子4多态性分析

摘    要:根据GenBank发表的人、鸡、大鼠雌激素受体(estrogen receptor,ESR)基因外显子4的序列设计1对引物,采用PCR SSCP技术分析ESR基因外显子4在高繁殖力绵羊品种(小尾寒羊和湖羊)和低繁殖力绵羊品种(特克塞尔、中国美利奴、考力代和杜泊)中的单核苷酸多态性,同时研究该基因对小尾寒羊高繁殖力的影响。结果表明,ESR基因的此对引物扩增片段在所检测的6个绵羊品种中均不存在PCR SSCP多态性,说明所检测的ESR基因外显子4序列比较保守,该区域可能不是影响绵羊高繁殖力的功能结构域。

Abstract:The estrogen receptor(ESR) gene was studied as a candidate gene for the high prolificacy of Small Tail Han sheep.A pair of primers was designed by sequence of exon 4 of ESR gene in human,chicken and rat.Single nucleotide polymorphisms(SNPs) of exon 4 of ESR gene were detected in both high prolificacy breeds(Small Tail Han and Hu sheep) and low prolificacy breeds(Texel,Chinese Merino,Corriedale and Dorper sheep) by polymerase chain reaction(PCR)-single strand conformation polymorphism(SSCP).No polymorphism was detected in the amplified region in the six sheep breeds tested.The results indicated that the sequence of exon 4 of ESR gene was rather conserved,and exon 4 may not be the functional domain that affects the high prolificacy in sheep.
