新陆早33号体细胞植株再生技术研究

摘    要:以模式品种Coker 312为对照研究新疆主栽陆地棉品种新陆早33号的体细胞胚胎发生和植株再生。研究表明,2个品种的下胚轴均能在0.05mg·L-1 2,4-D+0.1mg·L-1 KT的激素组合诱导下获得初生愈伤组织,然后在无激素诱导条件下获得胚性愈伤和体细胞胚。新陆早33号与Coker 312在MSB3培养基上最早体细胞胚胎发生时间分别为60d和87d,胚胎分化率分别为67.2%和61.4%。新陆早33号在180d获得根系发育良好的再生幼苗,而Coker 312需要210d。应用此法,成功获得新陆早33号的体细胞胚和再生植株,且其胚胎发生能力和再生效率均优于对照材料。

Abstract:Somatic embryogenesis and plant regeneration of Xiluzao 33 (Gossypium hirsutum L.) were studied, and compared with Coker 312. Calli were effectively produced on media with 0.05 mg·L-1 2, 4-D + 0.1 mg·L-1 KT hormones from hypocotyls. Embryogenic calli and somatic embryos were initiated on the media without any hormones. Somatic embryogenesis appeared after 60 d and 87 d on MSB3 medium, embryogenesis ratio was 67.2% and 61.4% respectively in Xinluzao 33 and Coker 312. It took about 180 d to obtain regenerated plants from somatic cells of Xinluzao 33, and about 210 d for Coker 312. Somatic embryos and regenerated plants have been successful obtained from Xinluzao 33 using the method, and its ability of embryogenesis and regeneration efficiency is better than Coker 312.
