天山雪莲种子萌发研究

摘    要:研究不同处理方式对天山雪莲种子萌发的影响,并优化种子萌发条件。20℃下采用不同浓度的Ca2+、Mg2+、Zn2+、Cu2+、PEG-4000、GA3分别浸泡天山雪莲种子48h,然后进行发芽试验,测定发芽率、发芽势、根长、发芽指数、活力指数,根据试验结果优选Ca2+、PEG-4000、GA3进行正交试验。结果:分别经Mg2+、Zn2+、Cu2+浸泡处理后,天山雪莲种子活力无显著提高;20℃时,Ca2+、PEG-4000、GA3浓度分别为20mmol/L、40g/L、200mg/L条件下浸泡种子48h,再进行发芽试验,发现较对照组发芽率提高45.4%,发芽势提高62.5%,根长提高12.5%,发芽指数提高73.9%,活力指数提高68.6%。结论:适宜浓度的Ca2+、PEG-4000、GA3及其复合处理能够显著提高种子性能。

Abstract:Objective: To investigate the influences of different treatments on germination of Saussurea involu-crate seeds and optimize the conditions of germination. Methods: After being respectively treated with Ca~(2+), Mg~(2+), Zn~(2+), Cu~(2+), PEG-4000 and GA_3 solutions in different concentrations for 48 hours at 20℃,S. involucra-ta seeds were incubated in an artificial bioclimatic chamber. The percentage of germination, germinative force, root length, germination index and vigor index were measured at the proper time. Based on the study result, Ca~(2+) ,PEG-4000 and GA_3 were chosen to the further optimized study with orthogonal test. Results: Mg~(2+), Zn~(2+), Cu~(2+) had no positive effects on vigor of S. involucrate seeds. However, compared with the control group, the percentage of germination, germinative force, root length, germinational index and vigor index increased by 45.4% ,62.5% ,12.5% ,73.9% ,68.6% ,respectively when the seeds were doused in a solution which con-centrations of Ca~(2+), PEG-4000, GA_3 were respectively 20 mmol/L,40 g/L,200 mg/L. Conclusion:Ca~(2+), PEG-4000, GA_3 and the combinative treatment with suitable concentration could significantly improve the germina-tional property of S. involucrata seeds.
