铜元素诱导烟草抗PVY~N相关生理生化及信号物质的研究

摘    要:通过气相色谱(GC)、高效液相色谱(HPLC)及生理生化指标的测定,研究了铜(Cu)元素诱导感染PVY~N的烟株信号物质的积累及对系统抗性的影响.结果表明:喷施Cu元素能延迟烟株显症和叶脉出现褐色坏死时间,降低发病程度,病增殖受刨抑制,烟株体内的与抗性相关物质如脯氨酸及可溶性糖含量升高,电解质的外渗减少.此外,Cu元素处理能诱导信号物质水杨酸和乙烯的积累,水杨酸含量在接种后15d达到最大量3.93μg·g~(-1),为接种对照的1.3倍;而乙烯释放量在3~9d内均高于接种对照,在12d时乙烯释放量显著低于接种对照,分析认为前期Cu元素处理后诱导了乙烯释放的增强,进行抗性信号的传递,因此,喷施Cu元素后可以诱导烟草植株产生抗病性,增强烟草对PVY~N侵染的抵抗力,并且诱导了相关抗性信号的传递.

Abstract:By the GC, HPLC, physiological and biochemical indices, the resistant and signal substances which induced by spraying copper against potato virus Y-vein necrosis strain (PVY~N) were studied. The results showed that when treated with copper at the concentration of 0.8mg·L~(-1), the symptom appearing was postponed, vein necrosis degree was lightened, the disease index and the content of virus sharply down. The content of prolinc and soluhle sugar in tobacco leaves were increased,and the leakage rate of electrolyte restrained. The content of salicylic acid (SA) reached the high peak at the 15th day after inoculation, which 1.3 times than that of inoculation control. The release of ethylene (ETH) was much higher than that of control in a short time. At the 12th day, ETH released from control was sharply increased, which was much higher than that of copper treatment, because the signal pathway in tobacco leaves was activated when treated with copper solution in early time. In conclusion spraying copper solution could induce tobacco plant resistance to PVY~N and activate the signal pathway. Those indicated that the induced resistance was promoted and the symptom was lighten.
