栎叶枇杷原生质体的分离与培养

摘    要:初步探讨了栎叶枇杷胚培养获得愈伤组织,从胚愈伤组织分离原生质体以及原生质体的早期培养方法,着重研究了酶混合液对栎叶枇杷原生质体分离和培养的效应.结果表明:以1%纤维素酶和2%果胶酶配合使用效果最好,得率和成活率分别为(9.8±0.03)×10 ̄6和96.0%.作为酶混合液中渗透压稳定剂的甘露醇浓度则以11%-12%为好.从合适的酶混合液中得到的原生质体培养已获得愈伤组织.

Abstract:The obtainment of callus from the cultured embryo in Eriobotrya prinodes Rehd.& Wils,the isolation of protoplasts from the callus and the culture of protoplasts,especially,the effects of enzyme rmxation on the isolation and culture of the protoplast,were dealt with in this paper.The results showed that the combination of1% cellulase with 2% pectose had the strongest effects on thhe isolation of protoplasts,gaining (9.8±0.03) ×10 ̄6/g fw and 96.0% the ratio of survival, and that 11%-12%mannitol was the optimum osmotic level.The isolated protplasts were cultured in the liquid medium and formed the newly callus.
