大豆叶片及种子内大豆花叶病毒(SMV)的生物与免疫测定方法的比較

摘    要:用生物测定(局部枯斑反应)、免疫吸附电镜(ISEM)、酶联免疫吸附试验(ELISA)三种方法对大豆叶片及种子内的SMV进行了定性定量测定。结果表明,三种方法均可用来检测叶片及种胚和子叶内的SMV,并可对其定量,结果高度吻合,相关系数达0.97以上。ELISA能测出稀释2430—7290倍病叶中的SMV,测提纯病毒的下限为70ng—20ng/ml,生物测定可测出稀释640—1280倍病叶中的SMV。种皮内含有非侵染性SMV,不能用生物方法测出,而需用免疫方法检测。


A COMPARISON BETWEEN BIOASSAY AND IMMUNE ASSAY FOR DETECTION OF SOYBEAN MOSAIC VIRUS IN SOYBEAN LEAVES AND SEEDS
Abstract:Local lesion assay, immunosorbent electron microscopy (ISEM)and enzyme-linked immuno-sorbent assay (ELISA) were compared for detecting soybean mosaic virus (SMV) in soybean leaves and seeds. The results obtained by these methods were highly correlated when detecting SMV in soybean leaves, embryos and cotyledons both qualitatively and quantitatively, with correlation coefficients as high as 0.97. However, the virus in soybean testae can not be detected by local lesion assay, which was found by immune methods.The sensitivity of ELISA and bioassay in detecting SMV in soybean leaf sap was 1:2430-1:7290, 1:640-1:1280 dilution respectively.The lowest concentration of purified SMV detected by ELISA was 70-20 ng/ml.
