蛋白质电泳在豌豆品种鉴定中的应用

摘    要:以25个豌豆品种为材料,分别用重蒸水2.5%NaCl、0.2%NaOH和75%乙醇提取其种子蛋白,利用连续型乳酸-聚丙烯酰胺凝胶电泳进行分离,结果表明:豌豆贮藏蛋白以水溶、盐溶和碱溶为主,醇溶蛋白甚微.三种主要蛋白在品种间存在不同程度的谱带差异,均可用于豌豆的品种鉴定,但以水溶和碱溶蛋白的谱带差异最大,是豌豆品种鉴定的首选蛋白.连续型乳酸-PAGE方法简便快速、分离效果好,是一种经济有效的方法.

Abstract:Seed proteins of 25 pea cultivars were respectively extracted using distilled H 2O, 2.5%NaCl, 0.2%NaOH and 75%alcohol. Based on coherent lactate polyacrylamide gel electrophoresis, the results showed that most of storage proteins were water-soluble, salt-soluble and alkali-soluble proteins. There is a little alcohol-soluble protein. All the three main proteins with polymorphism PAGE-banding patterns can be used in cultivars identification of pea, The salt-soluble and alkali-soluble proteins are more suitable to be used as biochemical marks of pea cultivars than salt-soluble. It also demonstrated that coherent lactate PAGE is a effective method in identification of pea cultivars, which is simple, rapid, inexpensive and has a good effect on protein isolation.
