摘    要:通过对传统双向电泳技术进行改进和优化,包括对锥栗果实蛋白不同提取方法的优化和不同pH值范围IPG胶条的比较等,以建立适于锥栗果实蛋白质组分析的双向电泳方法.结果表明,使用裂解液直接提取蛋白的方法所获得的双向电泳图谱稳定性和重复性较好,分辨率较高,经银染后可分辨出400多个蛋白点,其等电点主要分布于pH 4-7.

Abstract:The conventional two-dimensional electrophoresis technology was improved in order to develop an appropriate approach for proteomics of chinquapin fruits.The study was done by optimizing different protein extraction methods and comparing IPG strips at different pH values.With the improved protein extraction method,a stable,reproducible and high resolution 2-D electrophoresis map was obtained.After silver staining,more than 400 protein spots were detected,with their isoelectric points(pI) mainly at pH 4-7.
