摘    要:朊蛋白多肽PrP106-126可激活小胶质细胞并产生活性物质。探讨PrP106-126作用于BV-2小胶质细胞对NO生成状况影响,从酶学角度检测其作用机制。在细胞培养液中加入50μmol.L-1PrP106-126,培养48 h后检测培养液NO含量,采用实时定量RT-PCR检测细胞内iNOs和nNOs在mRNA表达水平。结果表明,PrP106-126显著提高细胞培养液中NO含量(9.34倍,P<0.01),且iNOs和nNOs表达水平均极显著提高(11.60倍和4.36倍,P<0.01),从而为解释朊病发生机制提供基础数据。

Abstract:PrP106-126 can increace cell viability of of microglia and the activated microglia can release cytokines,but the metabolic alterations of microglia remain poorly documented.A synthetic peptide corresponding to amino acid 106-126 of prion protein(PrP) has been shown to be toxic to neurons.In this study,VB-2 cell were used,the effects of PrP106-126 on contents of NO in BV-2 cells were investigated in vitro.The experiment was divided into three groups(Control,SCR,PrP106-126).The content of NO was significantly(P<0.01) increased when grown in media with PrP 106-126 at 48 h.The expression of iNOs and nNOs was examined at mRNA levels at 48 h.The results showed that exposure of PrP106-126 enhanced the level of NO(934%,P<0.01),the expression of iNOs(1160%,P<0.01)and nNOs(436%,P<0.01),and this may partially account for the pathogenesis of prion disease.
