转基因鲤与普通鲤鱼精巢结构和发育的对比观察

摘    要:通过光学显微镜对转大麻哈鱼生长激素基因鲤(以下简称转基因鲤)和鲤的精巢组织结构和发育进行比较观察。结果表明:转基因鲤和鲤鱼精巢组织结构基本上相同,都属于壶腹型精巢,精巢壁由两层膜构成,外层为腹膜,内层为白膜,二者精小叶的排列都不规则,每个小叶由许多精小囊组成。精子发生增殖、生长、成熟整个发育过程都在精小囊中进行,精子成熟后从精小囊进入小叶腔内。转基因鲤和鲤鱼精子的头部直径分别为2.60±0.026μm;2.58±0.024μm。性腺发育观察,7月份时在转基因鲤的精巢中精原细胞和精母细胞占多数;鲤鱼精巢中以精原细胞为主。8月份时转基因鲤精巢中精原细胞明显减少,而精母细胞开始增加,并有少量精子细胞和精子出现;鲤鱼精巢中也发现精子细胞和精子,9月份时转基因鲤和鲤鱼精巢中大多都为精子细胞和精子。通过对7~9三个月雄性发育程度作T检验,二者鱼的雄性性腺每个月发育程度有所不同但差异并不显著(P>0.05)。结果显示,转基因鲤和鲤鱼精巢组织结构基本上相同,转基因鲤与鲤鱼精巢发育都能达到性成熟,性腺发育程度无显著差异。

Abstract:The testis tissue construction and development of the transgenic crap that transform from the growth hormone of transgenosis salmon(hereinafter referred to as the genetically modified carp) and the carp are compared through the optical microscope.It turned out that the testis tissue construction of the genetically modified carp and the carp basically the same,they all belong to the ampullae type testis tissue,the spermary wall consists of two membranes,the outer is peritoneum,the inner is albuginea,the seminiferous lobules arrange of the two membranes are irregular,each of the seminiferous lobules consists of much seminal vesicle.The whole ontogenetic process of the proliferation,growth and mature is proceeding in the seminal vesicle,the sperm goes into the lobular lumen from the the seminal vesicle after matured.The head diameter of genetically modified carp and the crap sperm is 2.60±0.026 μm and 2.58±0.024 μm.The observation of the gonadal development found that the spermatogonium and the spermatocyte take the most part of the genetically modified carp testis tissue in July.The spermatogonium is the principal in the genetically modified carp testis tissue.During August,the spermatogonium in the genetically modified carp testis tissue is significantly reduced,In september,things in the genetically modified carp and common craps' testis are mainly sperm cells and sperm.Though T test of male craps,from July to September three months,development degree,the degree of genetically modified and common craps' gonadal development has some difference but the difference is not significantly(P>0.05).The experiment result shows that the genetically modified craps' testis structure is basically the same as common carps.the genetically modified craps and common craps' testis can both reach sexual maturity.The degree of gonadal development have no significant differences.
