新疆杏品种自交不亲和S-RNase基因特异PCR扩增引物的筛选

摘    要:[目的]筛选鉴定新疆杏自交不亲和相关S-RNase基因的有效引物.[方法]以新疆30个杏品种为试材,选用已报道的蔷薇科李属通用的5对引物组合进行S-RNase基因特异PCR扩增,并依据扩增系数和扩增成功率两个指标对扩增效果进行评价.[结果](1)5对引物组合对30个杏品种均扩出了条带,除了洛浦2号只扩出1条带之外,其余的29个品种都扩增出了两条带;(2)5对引物组合对新疆主栽杏的扩增效果差异较大,其中EM-PC2consFD+EM-PC3consRD的扩增成功率和扩增系数均为最高(86.67;, 95.00;),扩增出两条带的品种有25个;其次,引物组合PaConsⅡ-F+PaConsⅡ-R的扩增成功率达66.67;,扩增系数达83.33;,扩出两条带的品种有20个.[结论]5对引物中EM-PC2consFD+EM-PC3consRD对新疆杏品种S-RNase基因的鉴定效果最好.

Abstract:【Objective and Method】In order to select the primer for identifying S-RNase gene associated with self-incompatibility of apricots cultivated in Xinjiang,30 apricot cultivars were used for S-RNase specific PCR with 5 pairs of primer combinations which had been published for prunus consensus.【Result】The results showed:(1) The SRNase pecific bands were amplified in all the 30 cultivars by the 5 pairs of primers,and 29 cultivars except the "Luopu NO.2"showed two bands amplified.(2) The effect of amplification was significantly different among the 5 pairs of primers.The primer combination of EM-PC2consFD+EM-PC3consRD had the highest amplification coefficient(86.67%) and efficiency(95.00%),and two S-RNase specific bands were obtained in 25 cultivars.The amplification coefficient and efficiency of the primer combination PaConsⅡ-F+PaConsⅡ-R were 66.67% and 83.33% respectively,and two S-RNase specific bands were obtained in 20 cultivars.【Conclusion】Therefore,among the 5 pairs of primer combinations,EM-PC2consFD+EM-PC3consRD had the best identification effect for S-RNase gene for Xinjiang apricot cultivars.
