柞蚕抗菌肽D转基因酵母的培养及表达产物的分析

摘    要:采用碱金属法(醋酸锂盐),以大肠杆菌-酵母穿梭质粒表达载体在酵母中表达人工合成的柞蚕抗菌肽D基因。用电泳及抗菌肽活性检测法对表达产物进行分析。同时还研究了转基因酵母的发酵条件,如温度、pH及营养等对酵母表达的影响。试验结果表明转基因酵母表达产物为1.14mg/L培养液,杀菌活力为0.216单位。

Abstract:A synthetic antibacterial peptide gene(APG) from Chinese oak sikworm,Anther-aea pernyi was recombined with a shuttle plasmid pCL WA2 containing an alpha-factorpromotor and transferred into yeast AB103.The cells of yeast grew well under 18-30℃,pH 5.0 in YEPD medium.The expressed products were identified by electrophore-sis with detection of antibacterial activity.The expressed peptide has a molecular weightof 4000 daltons.The yield of product reached a level of 1.14mg/L YEPD and its activityunit is about 0.21 U.
