一个黑麦碱基因Sec-1表达缺失的抗白粉病新材料的鉴定和抗源分析

摘    要:小麦品系1002是经多年自交繁衍形成的稳定品系,对当前生产上流行的白粉菌生理小种表现苗期和成株期免疫抗性。为深入了解1002白粉病抗性的遗传基础,本研究对其抗性来源、遗传特性和细胞学背景进行了分析。结果表明,1002的白粉病抗性受一对显性基因控制,细胞核遗传,并能在感病品种中有效表达,与感病品种杂交F1代表现为免疫。1002是一个不含有中间偃麦草和簇毛麦遗传背景的黑麦碱基因Sec-1表达缺失的1BL/1RS易位系,其白粉病抗性基因与1BL/1RS染色体无关,与当前有效的白粉病抗性基因Pm37、Pm40、Pm43、PmCN17和PmL962并不相同。因此,推断1002携带一个新的抗白粉病基因。

Molecular Cytogenetic Identification of a Novel Powdery Mildew Resistance and 1BL/1RS Translocation Line with Secalin Absence
Abstract:Wheat 1002 is an optimization line that selected from germplasm resources we collected. The line 1002 exhibits immunity to the epidemic predominant races of powdery mildew. A F2 population and F3 families were developed from the cross of line 1002 and susceptible line MY11. Genetic analysis indicated that line 1002 possessed a dominant powdery mildew resistance gene. The allelic test and marker analysis showed that the gene was different from PmCN17, PmL962, Pm37, Pm40 and Pm43. In situ hybridization and A PAGE showed that line 1002 was a 1BL/1RS translocation line with secalin absence. PCR analysis by SCM9 which located on 1RS indicated that the resistant gene was not on 1BL/1RS chromosome.We also proved that line 1002 did not had H.villosa and Elytrigia intermedium background by using chromosome specific EST STS markers and SSR markers. The F1 generation, developed from the cross of line 1002 and cultivated varieties, exhibited immunity to powdery mildew, the resistant gene could be easily transferred to cultivated varieties. All these showed that line 1002 was a great and novel breeding material with powdery mildew resistance.
