向日葵食用蛋白的提取和SDS-PAGE电泳技术研究

摘    要:以向日葵种子为材料,采用SDS-PAGE电泳技术,以不同电泳条件对23种向日葵食用蛋白进行电泳谱带分析试验,研究不同电泳条件下向日葵食用蛋白的谱带变化,以选择出最佳的电泳条件。结果显示,最佳试验条件为:按照质量与体积比1∶3的比例用去离子水在4℃条件下30min提取水溶性蛋白。电泳时,在4℃条件下进行,凝胶板厚度1mm,分离胶浓度8%,浓缩胶浓度5%,电压300V,加样量控制在10μL。

Abstract:Taking sunflower seeds as materials, with different electrophoretic conditions, 23 kinds of edible sunflower protein electrophoresis band were analyzed by SDS-PAGE electrophoresis, in order to study the variations of electrophoresis protein bands of edible sunflower under different conditions and choose the best electrophoretic conditions. The results showed that optimum conditions were: according to the ratio of weight and volume 1∶3 , deionized water was used at 4 ℃ for 30 min to extract soluble proteins. Electrophoresis was done at 4 ℃ under the conditions of the gel thickness 1 mm, gel concentration of 8%, 5% stacking gel concentration, voltage 300 V, plus sample volume 10 μL.
