竹荪多糖的分离纯化和鉴定

摘    要:中国福建古田人工栽培的竹荪(Dictyophoraindusiata)子实休干品经热水提取,乙醇沉淀,蛋白法和Sevag法相结合去蛋白,DEAE-SephadexA-25和SephadexG-200柱纯化,得到竹荪多糖,命名为DI,用聚丙烯酰胺凝胶电泳和葡聚糖凝胶柱层析分析表明:该多糖为均一物质。竹荪多糖的分子量为144,000,经紫外扫描,元核酸和蛋白质的特征吸收峰,红外扫描表明有典型的多糖吸收峰。该多糖经纸层析和气相色谱分析表明:其单糖组成为L—岩藻糖、D—木糖、D—甘露糖、D—葡萄糖,摩尔比为0.16:0.24:1.00:1.08。

Abstract:A polysaccbaride ID was isolated from the fruit bodies of Dictyophora indusiata,itwas further purified by DBAE-Sephadex A-25 and Sephadex G-200,its homogeneity wasverified by gelfiltration chromatography and polyacrylamide gel electrophoresis.The com-ponents of DI were identified by paper and gas chromatography.It vas composed ofL-fucose,D-xylose,D-mann,D-glucose in the molar ratio of 0.16 : 0.24: 1.00: 1.08.Itsmolecular weight was estimated to be 144000.
