产生物胺乳酸菌的检测及产生物胺量的测定

摘    要:利用操作简便的产生物胺双层培养基检测法,对分离于发酵肉制品中的植物乳杆菌菌株进行了检测。利用高效液相色谱法对产生物胺的植物乳杆菌产生量进行测定,样品经浓度0.4mol/L的高氯酸提取后,用丹酰氯柱前衍生,流动相为乙腈和水,采用等梯度洗脱,流速为1.0mL/min,紫外检测器波长254nm,柱温40℃,进样量10μL。结果表明,采用该方法,两种主要生物胺组胺与酪胺能在10min内得到很好的分离,测得组胺质量浓度为211.03μg/mL,酪胺质量浓度为144.43μg/mL。

Abstract:Lactobacillus plantarum strain was isolated from the fermentation meat products and was detected simply by the method of producing biogenic amine double-layer medium detection. And then the producing content of biogenic Amine in Lactobacillus plantarum was also determined by HPLC. The results showed that,the two important biogenic amines(histamine and tyramine) can be separated well in 15 minutes,the concentration of histamine and tyramine were 211.03 μg/mL and 144.43 μg/mL.
