基于SSR分子标记的安徽黄山地区野生刺葡萄的遗传多样性分析

摘    要:利用8对SSR分子标记引物分析了安徽黄山地区40份野生刺葡萄资源的遗传多样性,结果表明:8对SSR引物扩增的等位基因数(Na)为36个,每个引物扩增等位基因数(Na)为2~10个,有效等位基因数(Ne)为1.285~5.378个;观测杂合度(Ho)为0.250~0.775,期望杂合度(He)为0.224~0.824;Nei’s多样性指数(H)为0.222~0.814;香农多样性指数(I)为0.417~1.884;多态性信息含量(PIC)为0.202~0.789。依据地理位置将黄山地区刺葡萄资源分为3个群体(POP1,POP2,POP3),3个群体之间遗传相似度为0.829~0.912,遗传距离为0.093~0.188,表明黄山地区刺葡萄资源的遗传差异较少。通过UPGMA方法对黄山地区40份刺葡萄和湖南、福建的3份刺葡萄进行聚类,在遗传相似系数为0.62时,黄山地区刺葡萄聚为一类,湖南、福建的刺葡萄聚为一类。这些结果表明,刺葡萄的基因流限制于短距离,黄山地区刺葡萄主要以有性繁殖为主。

Abstract:Eight pairs of microsatellite(SSR) molecular markers were used to analyze the genetic diversity of 40 individuals of wild Vitis davidii Fox collected from Huangshan region,Anhui province.The results showed that 36 alleles(Na) were amplified by the used 8 pairs of SSR primers,with a range of 2~10 alleles(Na) per primer,and the effective number of alleles(Ne) ranged from 1.285 to 5.378;the observed heterozygosity(Ho) varied from 0.250 to 0.775;the expected heterozygosity(He) was 0.224~0.824;Nei’s diversity index(H) was from 0.222 to 0.814;Shannon diversity index(I) ranged from 0.417 to 1.884,and the polymorphism information content(PIC) changed from 0.202 to 0.789.According to the geographic location,forty wild Vitis davidii resources in Huangshan region were divided into 3 groups(POP1,POP2 and POP3).Among these three groups,the genetic identity and genetic distance were 0.829~0.912 and 0.093~0.188,respectively,indicating a low level of genetic differentiation among populations of these three locations.The clustering dendrogram of 40 samples from Huangshan region and 3 samples from Hunan and Fujian was constructed by using UPGMA method,and it showed that the 43 individuals were divided into two groups when the genetic similarity coefficient was 0.62.The 3 samples from Hunan and Fujian were clustered as one group,and the others from Huangshan region were clustered as another group.These results indicated that the gene flow of Vitis davidii might be limited by the distance,and the principle reproductive mode of Vitis davidii in Huangshan region was sexual reproduction.
