中国李基因组DNA提取方法的优化

摘    要:3种常用的植物基因组DNA提取方法在中国李上的实验,结果表明:改良陈大明法提取的DNA纯度高,产率高。为满足植物样品较少时提取DNA的需要,将该法进一步优化的结果表明,不使用液氮研磨而是在低温条件下将组织捣碎的微量法同样提取出了产率高、纯度高的DNA,完全满足RAPD扩增的需要。裂解缓冲液中NaCl的浓度以1.1mol/L提取DNA的产率最高,且DNA的质量与其他浓度之间没有明显差异。

Abstract:Three common methods of genomic DNA isolated from plants were tested on Prunus salicina Lindl. The results showed that Modified Chen Method was the better way based on purity and quantity of genomic DNA. The modified micro-quantity method using tissue to be crushed under low temperature condition instead of grinding in the presence of liquid N2 was a useful way for DNA isolated from less amount plant material, and the quality of the genomic DNA was high and could be used for RAPD analysis. The results showed that the optimal concentration of NaCl in Lysis Buffer was 1.1 mol/L because the production of genomic DNA was higher and the quality was not apparently inferior to the others.
