禽腺病毒1型和4型六邻体蛋白抗原表位和密码子偏爱性分析

摘    要:为了研究禽腺病毒1型和4型(AAV-1和AAV-4)六邻体抗原表位和同义密码子的使用情况,运用生物学软件Antheprot 5.0和EMBOSS的CHIPS、CUSP程序分别对AAV-1和AAV-4六邻体进行了分析,并将分析结果与大肠埃希菌、酵母及人的密码子偏爱性进行比较.结果显示,AAV-1、AAV-4六邻体的抗原表位区都主要集中在前段、中段,推测具有型和群特异性表位,可作为诊断型和群的首选蛋白.AAV-1和AAV-4六邻体的Enc值分别为43.553和38.475,在编码密码子使用频率上都存在明显差异,与大肠埃希菌、酵母及人的密码子偏爱性也不同,其表达系统选择大肠埃希菌较为合适.

Abstract:In order to study the antigen epitope and condon preference of hexon protein of Avain adenovirus serotypes 1 and 4(AAV 1 and 4),their hexon sequences were analyzed by Antheprot 5.0 protein software.CHIPS and CUSPS programs of EMBOSS.The condon bias of them were compared with those of E.coli,yeast and human.The results showed that the anterior and middle part of the hexon protein contained of the majority of antigen epitopes that could be used as candidate type and group antigen for serological diagnosis.The ENc values of AAV-1 and AAV-4 were 43.553 and 38.475,respectively.There have distinct frequency differences in codon usage bias in AAV 1 and AAV 4.Condon preferences also were different from those of E.coli,yeast and human.E.coli system could be selected for AAV-1 and AAV-4 hexon gene expression.
