| 摘 要： | 根据GenBank上发表的猪传染性胃肠炎病毒(TGEV)基因序列,针对TGEV N基因全序列设计合成了一对引物,以TGEV疫苗株为模板,建立了检测TGEV的RT-PCR方法.应用该方法对TGEV疫苗株RNA进行扩增,获得与预期大小相符,长度为1 168 bp的特异性目的片段;测序结果与已报道的TGEV不同毒株序列的同源性为95%～97%;敏感性测定结果为可扩增到18 pg的TGEV N基因cDNA.结果表明,建立的RT-PCR方法对TGEV的检测敏感性高、特异性强,可用于TGEV感染的诊断和流行病学调查.

| Abstract: | A pair of primers were designed and synthesized based on the sequences of the TGEV genomes,and the RT-PCR method to detect the transmissible gastroenteritis virus(TGEV)was developed.The 1 168 bp specifical fragment was obtained from the TGEV vaccine strain RNA by this RT-PCR.The nucleotide homogeneity of the gene sequence of RT-PCR product was 95%-97% with that of other TGEV strains.The sensitivity of RT-PCR reached to 18 pg TGEV-N-cDNA.These results indicated that the RT-PCR method to detect TGEV was better specificity and higher sensibility and could apply to TGEV diagnoses and epidemiology investigation. |
