不同抗生素的浓度对百合鳞片再生的影响

摘    要:以兰州百合、OT型百合“木门”及东方百合“索邦”试管鳞茎的鳞片为外植体,在分别添加不同浓度头孢噻肟钠(Cef)、卡那霉素(Kan)和潮霉素(Hyg)的芽诱导培养基上进行培养,研究不同抗生素及其浓度对百合鳞片存活及再生的影响,筛选适宜的抗生素种类和浓度,为百合遗传转化和组培过程中细菌污染控制提供依据。结果表明,Cef对百合鳞片再生的影响较小,300 mg/L的Cef可用作农杆菌介导的遗传转化及组培快繁过程中细菌污染的脱菌剂;Kan对百合鳞片再生有较强的抑制作用,兰州百合最适宜的筛选浓度为40 mg/L,而“木门”和“索邦”适宜的筛选浓度为100 mg/L;Hyg对百合鳞片具有较强的致死作用,以Hyg为筛选标记进行兰州百合遗传转化时适宜的筛选浓度为25 mg/L,“木门”和“索邦”适宜的筛选浓度为35 mg/L。

Abstract:Using the vitro bulbs of Lilium davidii var.Unicolor,Conca D'or,Lilium oriental“Sorbonne”as explants cultured on the Murashige&Skoog medium with different concentrations of cefotaxime(Cef),kanamycin(Kan),hygromycin(Hyg)in this experient,the influences of the concentrations of different antibiotics on regeneration of lily bulbs were studied,and the appropriate concentrations of these antibiotics were chosen,which provided the theory basis for establishing the genetic transformation system of lily.The results showed that cefotaxime(Cef)had less effect on the regeneration of lily as a fungicide,and the concentration of 300 mg/L of cefotaxime(Cef)was feasible;when the concentration was up from 80 mg/L,the inhibiting effect of kanamycin(Kan)on lily bulbs was increasing gradually,and the optimal concentration of kanamycin was 40 mg/L for Lilium davidii var.unicolor and 100 mg/Lfor“Conca D'or”and Lilium oriental“Sorbonne”;the rate of bulbs browning dramatically increased above a concentration of 35 mg/L of hygromycin(Hyg),so chosing 25mg/L Hygfor Lilium davidii var.unicolor and 30 mg/Lfor“Conca D'or”and Lilium oriental“Sorbonne”was feasible.
