纤维素酶高产菌的分离纯化与产酶工艺优化

摘    要:从自然界采集到的各种样本中分离纯化纤维素酶产生菌,运用透明圈法进行初筛,将通过初筛的菌株进行摇瓶液体培养,测定各菌株的酶活,筛选出了一株酶活较高的纤维素酶产生菌.通过对该菌株在不同产酶条件(包括碳源、氮源、pH、温度、发酵时间)下羧甲基纤维素(CMC)酶活和滤纸酶活(FPA)的测定,发现其最佳产酶工艺条件为麸皮作碳源,黄豆粉作氮源,pH 6.5.在30℃下培养92 h.

Abstract:The cellulase producing strains were isolated from various samples collected from the nature,and screened by the methods of transparent zone and shaking culture.A strain with the highest cellulase enzyme activity was selected out.The fermentation conditions,including carbon sources,nitrogen sources,original pH,temperature and incubation time for this strain were optimized.And the CMCase and filter paper activity(FPA) were determined.The results showed that the optimum carbon and nitrogen source was bran and soybean flour respectively;and the best formation conditions for cellulase production were pH 6.5,cultivating for 92 h in 30 ℃.
