摘    要:用Design-Expert软件中的中心复合设计(Central composite design,CCD)模块,设计一组4因素5水平的试验,研究液料比、提取时间、乙醇体积分数和提取温度这4个因素对鲜桑椹中原花青素提取率的影响,构建具有良好预测性能的数学模型,通过响应曲面分析法确定鲜桑椹中原花青素的最优提取工艺条件.结果表明,回归模型为合适模型,优化提取工艺参数为:液料比22.5∶1,提取时间27.5 min,乙醇体积分数58.68%,提取温度66.25℃,在该条件下鲜桑葚原花青素的OD550nm理论值为8.22.经检验,所得工艺参数准确,可用于鲜桑椹中原花青素的提取生产.

Abstract:The central composite design model in Design-Expert software was used to design an assay with four factors at five levels in order to find the effects on the yield rate of procyanidins from fresh mulberry of four factors,such as liquid-solid ration,extraction time,ethanol concentration and extraction temperature.Based on it,a predictability mathematical model was constructed and the optimum conditions of procyanidins extration from fresh mulberry were determined using response surface analysis(RSA).The results showed that cubic mathematical model was the optimal one and the optimum extraction were as follow: liquid-solid ration was 22.5:1,extraction time was 27.5 min,ethanol concentration was 58.68%,extraction temperature was 66.25 ℃,and the theoretical yield OD550 nm was 8.22.The processing parameters in these experiments were accurate and could be applied in the mass extraction of procyanidins from fresh mulberry.
