芦笋ISSR引物筛选及遗传多样性分析

摘    要:采用ISSR技术对26份芦笋品种进行遗传多样性分析。以芦笋基因组DNA为模板,筛选出18条适用于芦笋的ISSR引物,并确定了它们的最佳退火温度。用这18条引物对收集自5个国家的26份芦笋品种进行ISSR分析,共获得扩增位点145个,其中多态性位点111个,多态性位点比例为76.6%。采用NTSYS2.10e软件进行相似系数分析,构建聚类树状图。在相似系数0.78处划线,可将样品划分为5个类群。

Primer Screening and Genetic Diversity Analysis of Asparagus by ISSR

Abstract:The genetic diversity of 26 asparagus varieties was analyzed by using ISSR.Based on the template of asparagus genomic DNA,18 ISSR primers were selected and their optimal annealing temperature were determined.26 asparagus cultivars collected from 5 countries were analyzed by ISSR with these 18 primers.A total of 145 DNA fragments were amplified by PCR,among which 111 fragments (76.6%) were polymorphic.The genetic similarity coefficient was analyzed,and a tree diagram was constructed by NTSYS2.10e software.UPGMA cluster analysis showed that these cultivars could be categorized into five groups according to coefficient 0.78.
