番茄同源四倍体离体诱导研究

摘    要:目的]利用组织培养技术与秋水仙素相结合进行番茄四倍体的离体诱导研究。方法]以番茄品种渝粉109为试验材料,在建立番茄植株高效离体再生体系的基础上,以带绿芽生长点的子叶愈伤组织为诱导材料,利用秋水仙素分别采用混培法和浸渍法进行番茄同源四倍体的离体诱导。结果]以带绿芽生长点的子叶愈伤组织为诱导材料,番茄同源四倍体离体诱导采用混培法,0.2%的秋水仙素处理48 h时获得了最高诱导率,为50.0%;浸渍法虽然诱导率较高,但对外植体的药害程度大。结论]通过混培法获得了较高的诱导率,为番茄品种改良和种质资源的创新与丰富提供了一条新的途径。

Study on in Viro Induction of Autotetraploid of Tomato(Lycopersicon esculentum Mill.)

Abstract:Objective] Study on in viro induction of autotetraploid of tomato was conducted by using the conbation of the tissue culture technology and the colchicine treatment.Method] With the tomato variety Yufen 109 as the tested material,based on the establishment of the tomato plant efficient in viro regeneration system,taking the cotyledons callus with the green bud growing point as the induction materials,the in viro induction of autotetraploid of tomato was conducted resp.by means of mixed culture method and immersion method with the colchicine.Result] When taking the cotyledons callus with the green bud growing point as the induction materials and in viro induction of autotetraploid of tomato was adopted by the mixed culture method,the treatment with 0.2% colchicines for 48 h could get highest induction rate,being 50%,while that by the immersion method got higher induction rate,but had a great harm on the explant.Conclusion] The higher induction rate was obtained by using the mixed culture method,which provided a new way for the improvement of variety and innovation and abundance of germplasm resources in the tomato.
