黑曲霉固体发酵产酸性蛋白酶条件优化

摘    要:对经诱变后黑曲霉固体发酵生产酸性蛋白酶的培养基组成及发酵条件进行优化。结果表明,该菌发酵产酶的最适培养基和条件为米糠20 g,碳源玉米粉60 g/kg,氮源硝酸铵5 g/kg,MnCl2 4 g/kg,料液比1.0∶1.5,调初始pH值7,以6mL/100g的接种量接种,在40℃、180 r/min下发酵72 h,摇瓶产酶达到1 120.0 U/mL,比基础培养基提高了近2.9倍。

Abstract:The medium composition and fermentation conditions were optimized for solid-state fermentation of Aspergillus niger.The experimental results showed that the optimum medium and conditions were as follows:rice bran 20g,6 g/kg Corn meal,ammonium nitrate nitrogen 5 g/kg,MnCl_2 4 g/kg,liquid ratio 1.0:1.5,adjusted initial pH7,6 mL/100g of the volume of cornvaccine inoculation,40℃、180 r/min fermentation time 72 h,in shake flask enzyme production rate reached 1120.0 U/mL,2.9 times more than that under base medium.
