摘    要:为研究透明颤菌血红蛋白基因vgb对黄原胶合成的影响,采用原生质体电转化的方法,将vgb与质粒pUC18构建的重组克隆pUC-LV,转入野油菜黄单胞杆菌中,并对转化菌和初发菌进行了PCR检测和摇瓶发酵试验。结果显示:vgb成功导入黄单胞杆菌,与初发菌相比,转化菌的黄原胶产量平均提高了15.91%,说明透明颤菌血红蛋白基因vgb的导入有利于黄原胶的合成。

Abstract:In order to study the effect of the Vitreoscilla hemoglobin gene(vgb) on Xanthan gum biosynthesis,the recombinant clone pUC-LV,which was constructed by cloning vgb to the vector pUC18,was transferred into Xanthomonas campestris by the protoplast electro-transformation in the study.The PCR amplification and the shake-flask fermentation were done between the wild type strain and the transformants.The results showed that vgb had been transferred into Xanthomonas campestris successfully,and the yield of transformants had increased by 15.91% on average than the wild type strain in fermentation.These results demonstrated that vgb played a role in enhancing Xanthan production.
