草樱花组培快繁技术的研究

摘    要:以草樱花为材料,研究了激素配比、蔗糖、培养条件等组培苗工厂化生产关键技术。结果表明:采用MS+6-BA0 5mg/L+IBA0 05mg/L(或2,4-D0 25mg/L)+蔗糖30g/L用于分化培养,1/4MS+NAA0 5mg/L+蔗糖10g/L用于生根培养,光强1500~2000lx,光照时间每天12~14h条件下组培苗增殖系数最大,有效苗最多,生根率高及生长速度快,移栽成活率90%以上。采用管外扦插成活率80%以上。建立了草樱花工厂化生产工艺及试管苗培养标准,为其植物组培苗繁殖提供了理论依据。

Abstract:Culturing the stem of P. subulata L. in vitro, the key techniques including medium,hormones,sucrose,culture conditions et al. were studied.The results showed that if there was MS+ 6-BA 0.5mg/L+ IBA 0.05mg/L(or 2,4-D 0.25mg/L)+ sucrose 30g/L for mutiplicating, 1/4MS +NAA 0.5mg/L+sucrose 10g/L for rooting,light intensity sustaining under 1500~2000lx and illuminated for 12~14h/d, the coefficient of multiplication,quantity of effective plantlets and rooting rate were best.The survival rate of transplantation was above (90%.)More than 80% of plantlets survived by using outer test-tube cuttage for rooting.Based on the study,the industrializing technology of micropagation of P.subulata L. by in vitro culture and the culture standard which improves a theoretical basic for micropagation of other plants were summarized.
