金线莲组培快繁技术体系的研究

摘    要:通过设置不同的试验来优化金线莲的组培条件,结果表明:金线莲外植体消毒处理以75%乙醇(浸泡30 s)+0.1%HgCl(浸泡8 min)的效果较好,诱导芽培养基以MS+BA 2.0 mg/L+NAA 0.3 mg/L的处理效果最优,芽丛生增殖培养基以MS+BA 1.5 mg/L+NAA 0.5 mg/L的效果最优,在增殖培养基中添加20%香蕉泥对芽苗的增殖及壮苗培养均有极大的促进作用。在芽体分化培养过程中,在MS+BA 0.05 mg/L+NAA 0.5 mg/L的培养基中,接种密度为每瓶10颗和800lx的光照处理效果较为理想。

Abstract:Different tests were conducted to optimize the tissue culture conditions of Anoectochilus roxburghii.The results showed that 75% ethanol(soaking for 30 seconds) + 0.1% HgCl(soaking for 8 minutes) had better disinfection effect on the explants.The best medium for the bud inducement was MS+BA 2.0 mg/L+NAA 0.3 mg/L,while the best medium for the bud proliferation was MS+BA 1.5 mg/L+NAA 0.5 mg/L.Addition of 20% banana puree in the proliferation medium could improve bud proliferation and culture of strong seedlings.In the culture process of bud differentiation and the culture medium of MS+BA 0.05 mg/L+NAA 0.5 mg/L,the inoculum density of 10 plants per bottle and the light treatment of 800 lx could produce ideal effects.
