乌拉尔甘草离体根尖培养方法的建立

摘    要:建立甘草的离体根尖培养体系有助于研究甘草的根系营养、形态建成以及某些次生代谢产物的合成、代谢等。为建立乌拉尔甘草的离体根尖培养体系,对影响乌拉尔甘草离体根尖生长的培养基、激素以及维生素等因素进行了研究,并对其中的药用有效成分进行分析。结果表明:在培养基中附加0.10mg/L的IBA,可诱导乌拉尔甘草离体根尖伸长并产生较多的侧根;B,大量元素+B5微量元素和1/3MS大量元素+1/3MS微量元素适宜乌拉尔甘草离体根系的生长。表现为主根和侧根发达而均匀,具有较强的生长活力;培养基中附加0.50mg/L的VB1可显著促进乌拉尔甘草离体根系鲜重的增加;液体悬浮培养适于乌拉尔甘草离体根尖的培养。在悬浮培养条件下甘草离体根中甘草总黄酮物质的含量为0.75%,但没有检测到甘草酸的产生。

Abstract:In Vitro root tip culture of Glycyrrhiza uralensis Fisch. is an important system for study on root nutrition, morphological development and metabolism of some secondary components. In order to establish In Vitro root tip culture method of licorice, some influence factors including phytohormone, culture medium, vitamin and holding-medium, were studied in this paper. The results showed that IBA concentration of 0.1 mg/ L could induce root tip to lengthen and produce more adventitious roots. Macroelement and microelement of B5 and 1/3 MS basal medium, VB1 concentration of 0.50 mg/L and liquid suspension culture were more suitable condition for root tip culture of licorice. The total flavonoids content of 0.75% was obtained in licorice in vitro root cultured under screened optimum liquid suspension condition, but glycyhhizic acid was not detected.
