文心兰侧芽诱导丛生芽技术研究

摘    要:以文心兰侧芽为外植体,研究侧芽采前处理、消毒次数、培养基中不同椰子汁含量配比对丛生芽诱导的影响。结果表明,采用在培养基中添加天然植物体,以幼嫩的椰子汁本身所含的微量激素和植物细胞分裂所需的关键诱导因子代替人工合成激素,对以文心兰侧芽为外植体进行丛生芽的诱导效果明显;以文心兰侧芽为外植体,通过采前5~7d对植株的控水和喷施抗菌剂相结合的方法处理后,可避免病菌感染;经过连续2次0.15%的升汞溶液灭菌(其中消毒时间为第1次7~8min,第2次30~50s),消毒效果最好;以(1/3大量元素)MS+150mL椰子汁+50g/L番茄+20g糖+7g琼脂,pH为5.5~5.8的培养基中,黑暗环境条件下经过30d左右的培养,即可见有丛生芽的形成。

Abstract:Oncidium lateral buds used as explants were treated and cultured to observe the effect of pretreatment, disinfection frequency, and different levels of coconut water in the medium on inducing of multiple shoot clumps. The Oncidium lateral buds were cut 5-7 days after the plants were controlled by irrigation and sprayed with antimicrobial agents to avoid pathogenic infection; the buds then were disinfected 2 times with mercury solution at the concentration of 0.15% (initial disinfection for 7 to 8 minutes and the second disinfection for 30-50 seconds); visible multiple shoot clumps were induced from the explants which were cultured for about 30 days under the dark conditions on the cultural medium of MS containing 1/3 macro-elements +150 ml coconut water +50 g/l tomato +20 g sugar + 7 g agar, with the pH 5.5-5.8.
