小麦-冰草异源衍生系籽粒蛋白质特性分析

摘    要:为了研究小麦-冰草异源衍生系的蛋白质品质性状,以大面积推广的6个小麦品种为对照,采用国际(ICC)标准和常规分析方法,对16个小麦-冰草异源衍生系的蛋白质组分、湿面筋含量、SDS沉淀值、谷蛋白溶涨指数(SIG)以及高分子量谷蛋白亚基(HMW-GS)组成等蛋白质品质特性进行了系统分析.结果表明,小麦-冰草异源衍生系平均蛋白质含量为11.60%,略高于对照品种的平均值(11.31%);蛋白质组分中,清蛋白、谷蛋白的平均含量分别为26.02%和44.15%,略高于对照品种,球蛋白、醇溶蛋白的平均含量分别为5.96%和24.30%,略低于对照品种;湿面筋含量、SIG的平均值显著高于对照品种;SDS沉淀值的平均值略高于对照品种;HMW-GS电泳分析显示,含有7 8优质亚基、亚基总评分为8分的小麦-冰草异源衍生系占81.25%.16个衍生系中,1-1-7、10-36、10-78、20-1-1-2和20-1-1-2-4的蛋白质含量、湿面筋含量、SDS沉淀值、SIG值均高于对照品种的平均值,HMW-GS组成均为1/7 8/2 12,可作为小麦优质蛋白质基因的供体.

Abstract:To study the protein property in kernels of Triticum aestivum-Agropyron cristatum heterogeneous derivative lines(called "derivative lines" for short in following),the protein compositions,wet gluten contents,SDS sedimentation values,swelling indexes of glutenin(SIG),HMW-GS compositions of 16 derivative lines were studied systematically by routine analysis and the ICC Standard using six wheat cultivars as the control.The results indicted that the protein contents of the derivative lines averaged 11.60% and slightly higher than that of the controls(CK).Among the average protein component contents(percentage of each protein component content to total protein content) of derivative lines,albumin is 26.02% and slightly higher than CK;globulin is 5.96% and slightly lower than CK,gliadin is 24.30% and significantly lower than CK;and glutenin is 44.15% and significantly higher than CK.The wet gluten contents and SIG of the derivative lines are significantly higher than CK;the average of SDS sedimentation values is slightly higher than CK;HMW-GS analysis by electrophoresis showed that 81.25% of the derivative lines had the good subunits combination 7 8.Derivative lines 1-1-7,10-36,10-78,20-1-1-2-2 and 20-1-1-2-4 had higher protein content,wet gluten contents,SDS sedimentation values and SIG,and their HMW-GS subunits compositions are 1/7 8/2 12,and they could be used as the donors of quality-protein genes for wheat.
