蜈蚣藻多糖纯化及抗氧化作用的研究

摘    要:采用热水浸提法提取蜈蚣藻多糖,通过对超氧阴离子(·O2-)、羟基自由基(·OH)清除效果的研究,评价了蜈蚣藻多糖的体外抗氧化活性,以及对蛋白脱除率和多糖损失率的分析,比较了TCA法和Sevage法脱蛋白的效果,利用Sephadex G-150柱纯化蜈蚣藻多糖,并分析了其部分理化性质和相对分子质量分布.结果表明,蜈蚣藻多糖对·OH清除率的IC50为4.62 mg/mL,对·O2-清除率的IC50为0.62mg/mL,具有体外抗氧化效果;Sevage法脱蛋白效果优于TCA法,蛋白脱除率达85.55%,多糖损失率为15.93%;蜈蚣藻多糖是一种以多糖为主、含有糖蛋白的复合物,其相对分子量的分布范围在3.5×105 ~1.2 ×105 Da.

Abstract:In this paper, authors assess vitro antioxidation by studying cleaning effect of ·O2- and · OH and compare trichloroacetic acid (TCA) method with Sevage method by analyzing deproteinization rate and poly- saccharides loss rate. Then the authors purify polysaccharides and determine the distribution of molecular weight through using glucan gel G - 150 chromatography column, furthermore analyze some of its physical and chemical properties. The results indicate that polysaccharides from Grateloupia filicin has antioxidant activity, IC50 on scavenging ·OH is 4. 62 mg/mL, IC50 on scavenging ·O2- is 0. 62 mg/mL. On effect of deprotein- ization, Sevage method is superior to TCA method, deproteinization rate is 85.55% and polysaccharides loss rate is 15.93%. Polysaccharides from Grateloupia saccharides and glycoprotein, and the distribution filicin is a kind of compounds with major constituent poly- of molecular weight is between 3.5 - 105 and 1.2 ×10^5 Da
