菊花遗传转化受体系统的研究

摘    要:探讨了不同抗生素对菊花品种“玉人面”叶片不定芽分化和无菌苗生长的影响,并对羧卞青霉素、头孢霉素的抑菌效果及菌株LBA4404、EHA105对菊花的侵染性进行了研究,建立了“玉人面”的遗传转化受体系统。结果表明:叶片不定芽再生、无菌苗生根筛选的抗生素和临界耐受浓度分别为10 mg/L的G418、20 mg/L的卡那霉素;250 mg/L的抑菌抗生素不影响叶片外植体不定芽再生,两种抑菌抗生素均影响菊花苗的生长,总体规律是随抗生素浓度的递增,菊花苗生根数量递增,根长、苗高递减;两种抑菌抗生素均能很好的抑制农杆菌的生长并对“玉人面”有一定的侵染性。

Abstract:Effects of antibiotics on shoot regeneration and reducing root onChrysanthemumcultivars 'Yu Ren Mian'was studied.At the same time,the influence of Cefotaxime and Carbenicillin on growth ofAgrobacterium tumefaciensand infection ability of LBA4404、EHA105 on chrysanthemum were discussed.The transformation receptor system for Chrysanthemumwere established.The results showed that the critical concentration for shoot regeneration and reduc-ing root forChrysanthemumwere 10 mg/LG418 and 20 mg/Lkanamycin in respectively.The**re had no impact on shoot regeneration by 250 mg/L Cefotaxime or Carbenicillin.The growth ofchrysanthemumwas affected by Cefo-taxime or Carbenicillin.The number of root increased,while the length of root and height of chrysanthemum de-creased as the antibiotics concentration increased;Growth ofAgrobacterium tumefacienswere restrained by Cefotaxime or Carbenicillin and 'Yu Ren Mian'could be infected by LBA4404 and EHA105.
