应用竞争性酶联免疫吸附试验检测鸡产蛋下降综合症抗体

用鸡产蛋下降综合症(Egg Drop Syndrome-76,EDS-76)灭活油佐剂苗,免疫30周龄产蛋鸡,获取高免血清。提取特异性免疫球蛋白后,用辣根过氧化酶标记,制备成酶标抗体,建立了竞争性酶联免疫吸附试验。该法与血凝抑制试验的平行测定的比较结果为,前者的阳性检出率为71.8%,而后者为59.0%,所以前者明显地优于后者。而且该试验敏感性高,特异性强,具有快速、准确的优点,适用于大规模的检测和检疫。

DETECTION OF THE ANTIBODY OF AVIAN EGG DROP SYNDROME-76 BY USING THE COMPETITION ENZYME LINKED IMMUNOSORBENT ASSAY

The 30 weeks-age hens were immunized with the inactivated oil-emulsion Egg Drop Synd- rome-76(EDS-76)virus vaccine,and ether to yield the hyperimmune serum.The immune globulin was obtained from this serum by gel filtration in Sephadex G-50,and labeled with the horse-radise peroxidase(HRP).then,the competition enzyme linked immunosorbent assay (ELISA)was established.Comparison of two detection methods used to detect some EDS-76 position sera,the competition ELISA and the hemagglutination inhibition(HI)test,demonstra- ted that the former gave the 71.8 per cent positive detection rate,and the later had the 59.0 per cent positive detection rate.This competition ELISA not only had higher sensitivity and better speciality,but also had rapid and accurate advantages.
