红心杉花粉萌发率的测定及其储藏条件研究

摘    要:以陈山红心杉花粉为材料,对其花粉形态、离体培养萌发率以及不同储藏温度下的生活力进行了研究,结果表明:陈山红心杉新鲜花粉的生活力达到96%以上;花粉在恒温25℃,15%蔗糖+10 mg/L硼酸+1%琼脂的固体培养基上培养6 h时萌发率达到83.3%。不同储藏温度对花粉的生活力有较大影响,常温储藏时花粉生活力丧失较快,一周后显著下降,一个月后下降到59.9%,储藏90 d后活力仅有4.9%;花粉在-70℃下储藏1 a后生活力下降为40.8%。因此,花粉的长期储藏应在-70℃冷冻条件下效果最好,而在实践中,最适宜授粉的时期应在花粉散粉后的一周之内。

Abstract:The pollens shape,pollen germination rate in vitro and vitality of germination percentage of Cunninghamia lanceolata under different storage conditions were studied.The results showed that fresh pollen vitality of C.lanceolata reaches over 96%.Germination rate topped in the culture medium of 15% sucrose+10 mg/L boric acid+1% agar under 25 ℃ after 6 hours,reaching 83.3%.There were big affections to vitality of pollen under different storage conditions.Room temperature was not good for pollen preservation of C.lanceolata,while both-20 ℃ and 4 ℃ conduce to it.Significantly decreased after a week,a month later dropped to 59.9%,after 90 d of storage vitality only 4.9%.After dry storage at-70 ℃ for one year,the germination rate of C.lanceolata was 40.8%.For long time preservation(1 year),it was an overwhelming conclusion that-70 ℃ was better than 4 ℃ and-20 ℃.The most suitable pollination date was one week after blooms through practice.
