类胡萝卜素合成酶基因LycB导入番茄的研究

摘    要:以4个番茄品种的子叶为受体,以LycB为目的基因,应用根癌农杆菌介导法对番茄进行了遗传转化,经愈伤诱导、芽诱导、生根培养和潮霉素选择培养,获得的抗性植株经PCR和PCR-Southern分析鉴定,初步证明外源基因LycB已整合到番茄的基因组中。

Abstract:The tomato genetic transformation system was established by cocuhivated cotyledons of four varieties of tomato with agrobacterium tumefaciens EHA 101 including the plasmid pEbisLycBHyg. Then LycB gene can be transformed into tomato via agrobacterium tumefaciens. Regeneration plants of tomato were obtained by callus inducing, bud inducing, striking root cultivation and hygromycin screening in selective condition. PCR and PCR-Southern analysis of these resistant plants proved that LycB has been transferred into genome of tomato. The factors affecting genetic transformation were also investigated.
