卷丹百合3种主要病毒脱毒方法研究

为了获得无病毒的优质种苗,以感染CMV、LSV和LRV的卷丹百合珠芽为试材,采用热处理、茎尖培养结合添加病毒唑的方法,运用RT-PCR方法检测病毒,探讨最适宜的脱毒方法。结果表明:对于CMV,仅通过茎尖培养,剥取0.1~0.3 mm大小茎尖培养脱毒率可达100%,但成活率仅为45%;对于LSV,38℃高温热处理20 d后,剥取0.1~0.3 mm茎尖培养,脱毒效果最佳,可达90%以上;对于LRV,38℃高温热处理20 d后,剥取0.4~0.6 mm茎尖培养,添加10 mg/L病毒唑可使脱毒率达90%以上。综合3种病毒的脱毒率和成活率,以38℃高温热处理20 d后,剥取0.4~0.6 mm茎尖培养,添加10 mg/L病毒唑,成活率较高,脱毒效果最好。

Detoxification Methods of Three Viruses of Lilium lancifolium

In order to obtain high quality seedlings without virus, the tigerLilium lancifolium three main virus detoxiifcation methods were studied. Taking CMV infection, LSV and LRV tiger Lilium lancifolium bulbil as experimental materials, the heat treatment, stem tip cultivation method of adding ribavirin combination, in the process of test using RT-PCR method to detect virus, discussed the optimum method of detoxification. Research results showed that: for CMV, only through cultivation stem tip, wring 0.1-0.3 mm size stem tip seedling cultivation rate can reach 100%, but the survival rate is only 45%; For LSV, heat treatment(38℃) in 20 days, wring 0.1-0.3 mm meristem-tip culture, detoxiifcation effect was the best, reached more than 90%; For LRV, heat treatment(38℃) in 20 days, wring 0.4-0.6 mm meristem-tip culture, add 10 mg/L ribavirin the detoxication rate reached above 90%. Comprehensive seedling rate and survival rate of the three kinds of virus, with heat treatment(38℃) after 20 days, wring 0.4-0.6 mm meristem-tip culture, add 10 mg/L ribavirin, high survival rate, detoxiifcation effect was the best.
